The default LMB option must be enough for most the cases. The RMB allow you increase the "height" value and play with X and Y inversion.
The Y inversion can be useful if you want create an normal map compatible with DirectX: -Y means DirectX; +Y means OpenGL. Wings3d default is OpenGL.
If you invert X and Y together you will get the whole map inverted. So, what was up will be down and vice versa.
You can use an color image as input. It will be automatically converted to grayscale based on its luminance. But, sometimes you will get better results if you extract the bump/height map from it by using in an image editor software since you have more options.
